Les microARN et leur potentiel thérapeutique en cancérologie : le point en 2019

Abstract : Since their discovery in the 1990s, microRNAs (miRNAs) have fascinated scientists with their ability to regulate a broad spectrum of genes post-transcriptionally and finely, and to control almost all cellular processes. MiRNAs are key mediators of gene reprogramming and cellular functions under physiological and pathological conditions, particularly in cancer. This mini-review summarizes the steps in miRNA biogenesis and regulation of mRNA translation and stability. We remind the molecular matching rules between a miRNA and its target and evoke the dense and complex molecular network made up of thousands of interactions between genes and miRNAs and highlighted by the phenomenal advances in bioinformatics and RNA sequencing in recent years. Finally, we highlight their role in carcinogenesis and their therapeutic potential as anti-cancer agents, giving some examples of miRNAs evaluated in clinical trials. In conclusion, we stress the importance of continuing research efforts in miRNA field, particularly for the vectorization of small RNAs, because despite the obstacles to be overcome, they remain major and promising candidates in cancer treatment.
