Application au mésothéliome, le cancer de l'amiante

Abstract : Polymeric nanoparticle for treating cancers: application in mesothelioma, the asbestos-related cancer This article presents the selective tumour delivery of histone deacetylases inhibitors using a polymeric nanoparticle platform produced by ring-opening metathesis polymerization (ROMP) of azido-polyethylene oxide-norbornene macromonomers functionalized using click chemistry. In our models of mesothelioma, a highly selective accumulation of the particles in the tumour was obtained. The release of the drugs involved a pH-responsive polymer conjugate leading to 80% reduction of tumor weight for the best compound without toxicity. This work demonstrates that the use of theranostic nanovectors leads to an optimized delivery of epigenetic inhibitors in tumours and then, improving their anti-tumour properties in vivo.
