, Autoreactive IgE included in circulating immune complexes (ICs) induce cross-linking of Fc?RI and engagement of ITAM-mediated signaling. This leads to synthesis and secretion of IL-4 by basophils Autoreactive IgG also included in these ICs may engage Fc?RIIa and Fc?RIIb, with a ratio activation/ inhibition depending of the amount of IgG. Autoreactive IgE and IgE included in circulating ICs act in synergy, and bind, respectively, to Fc?RI and Fc?RIIa on plasmacytoid dendritic cells (pDCs) This synergic mechanism overloads the inhibitory signal mediated through Fc?RIIb engagement. Subsequent delivery of ICs containing dsDNA leads to TLR9 sensing, phosphorylation of the transcription factor IRF7 and production of type I interferon (IFN) and other inflammatory cytokines.
