Résumé

Fragment-Based Drug Discovery consist of the development of new drugs from fragments (small organic molecules of weak affinity for the target protein) and requires screening methods suitable for the detection of weak affinities (μM-mM). Recently, Weak Affinity Chromatography has been shown to be an interesting alternative approach for fragment screening and consists of immobilizing the target protein on a chromatographic support and then measuring the fragments retention, directly linked to their affinity for the target. To minimize the protein consumption and allow the study of targets difficult to produce, we decided to develop this approach at a miniaturized scale. In practice, monoliths were in-situ synthesized in 75 µm i.d. capillaries and the target model protein was in-situ grafted. Columns were evaluated using frontal analysis with ligands of known affinity for a model protein. Among different combinations, best results were achieved with a streptavidin modified organic monolith which allows the fast immobilization of biotinylated target protein (through the strong streptavidin-biotin interaction). We then focused on a membrane protein, the adenosine A2A receptor (AA2AR), embedded in a biotinylated nanodiscs then grafted on streptavidin columns, consuming less than 1 µg of protein per column. Affinities down to hundreds of µM were quantified, confirming protein activity after immobilization. Le Fragment-Based Drug Discovery consiste à développer de nouveaux médicaments à partir de fragments (petites molécules organiques de faible affinité pour une cible thérapeutique) et requiert des méthodes de criblage capables de détecter de faibles affinités (μM-mM). Récemment, la Chromatographie de Faible Affinité s’est avérée être une alternative intéressante aux méthodes de criblage de référence et consiste à immobiliser la protéine cible sur un support chromatographique puis à mesurer la rétention des fragments, directement liée à leur affinité pour la cible. Pour limiter au maximum la consommation en protéine et permettre l’étude de cibles difficiles à produire, nous avons décidé de développer cette approche à échelle miniaturisée. En pratique, un monolithe est synthétisé in-situ dans un capillaire de 75 µm et la protéine est également greffée in-situ. Les colonnes sont évaluées par analyse frontale avec des ligands d’affinité connue pour une protéine modèle. Parmi les différentes combinaisons testées, les meilleurs résultats ont été obtenus avec un monolithe organique fonctionnalisé streptavidine, permettant l’immobilisation rapide de la protéine cible biotinylée (interaction streptavidine-biotine). Nous avons alors travaillé sur une protéine membranaire, le récepteur A2A de l’adénosine, inséré dans un nanodisque biotinylé puis greffé sur des colonnes streptavidine en consommant moins d’1 µg de protéine/colonne. Des affinités jusqu’à quelques centaines de µM ont pu être détectées et quantifiées, confirmant l’activité de la protéine après immobilisation. L’identification de ligands parmi des fragments a permis d’illustrer le potentiel de cette technique pour le FBDD
